Metabolic modulators Metabolic modulators such as 2-deoxy-D-glucose (2-DG), which acts as an inhibitor of glycolysis, have been shown to reduce the expression of terminal exhaustion markers such as Tim-3 and LAG-3 in T cells while increasing T cell persistence, suggesting that the strategy of short-term inhibition of glycolysis preserves T cell exhaustion without compromising their effector functions and shows promise as a potential metabolic intervention strategy to maintain the antitumor activity of T cells [25]
